What should I do with the weeds in my interlock?

You should get rid of them.
Here is what you should do.
Assess the size of the weeds. If they are large, you may want to pull them by hand. Or destroy them with a blow torch.
If they are small, use a pressure washer to wash them out. You will also be washing out the soil the are growing in.
Then go through everything and make sure you got all the weeds out.
After you think you have the weeds out, spray the patio off to make sure it is clean of dirt and polymeric sand residue. (Polymeric sand is the concrete-like sand that your sweep into the joints of your interlock pavement. It is called polymeric sand because it is sand with cement polymers in it.)
Now go through everything again, but this time with a container of herbicide to kill all the weeds that are hiding in the joints. Have no mercy!
After this, wait a day for the herbicide to dry and have effect.
Then put new polymeric sand in the joints.
